Application Strategies on Volume Electron Microscopy

Volumatric electron microscopy imaging encompasses TEM-based serial section imaging and serial section electron tomography, as well as SEM-based technologies such as array tomography, focused ion beam-SEM (FIB-SEM), and serial block face-SEM (SBF-SEM). As a fee-for-service microscopy core facility, our goal is to design tailored approaches that address specific research questions related to ultrastructural morphology and molecule localization within the complex environments of cells and tissues across diverse biological sample types. The successful of a project depends on selecting the appropriate approaches, careful experimental design, optimized sample preparation, and high-quality microscopy imaging. In this talk, I will present case studies of successful projects to highlight key strategies in experimental design, focusing on the critical aspects of serial section TEM, serial section electron tomography and SBF-SEM - from sample preparation to imaging considerations.
